Abstract:
The current application is in response to the request by the AFI Moderating Panel (June 2000) to submit an application for an exploratory study to develop microsatellites and test for population structuring in Antarctic fishes as a basis for a subsequent full three-year AFI bid. The emphasis is thus on technical development and initial tests to demonstrate the feasibility of the approach within the Antarctic context, though the ultimate aim is to explore gene flow in relation to oceanography and diversity in life-history. Specifically, we will (1) develop microsatellite primers for two species of Antarctic fish, Champsocephalus gunnari and Notothenia rossii; (2) undertake a pilot analysis of the extent and patterns of genetic differentiation in populations of one species (C. gunnari); (3) perform initial computer simulations testing the utility of mixed stock analysis (MSA) for subsequent determination of larval origin.
