Abstract:
We have identified a polymorphism within the timeless (tim) clock gene of D. melanogaster that generates two TIM isoforms, and reveals a robust clinal distribution within Europe. We will study the functional significance of this polymorphism, and the well-known Thr-Gly cline within the PER clock protein, studying gene frequencies at different altitudes in natural populations. We will investigate the effects of temperature and photoperiod on transgenic flies carrying the two types of TIM variants, particularly the photoperiodic response of diapause in females, which appears to be determined by TIM, and also determine the properties of the two TIM protein isoforms when challenged with environmental change. Finally we shall examine the evolutionary and phylogenetic history of the TIM variants.
