Abstract:
This proposal will study the impact of a polychlorinated biphenyl (PCB 126) on immunoregulatory mechanisms in fish to gain an in-depth understanding of how PCB-induced lesions within the immune system are brought about. Specifically we will: 1) determine the impact of feeding PCB 126 on the expression of cytokines that initiate and regulate immune responses, focussing on interleukin-1beta, tumor necrosis factor alpha and IL-8, 2) determine the impact of feeding PCB on cytokine recptor expression (IL-1RI, TNFR1 and IL-8R), and key intracellular signalling molecules used by cytokine receptors, 3) confirm whether any changes in cytokine gene expression due to PCB exposure are seen during infection, with potential for affecting disease resistance, 4) determine the impact of a secondary stressor on cytokine/cytokine receptor expression in PCB exposed fish and 5) attempt to clone the trout leptin gene to allow functional analysis of this important link between the stress response, energy balance and the immune system in PCB exposed fish.
