- Abstract:
- A survey of bacterial phenotypic resistance to surface-active quaternary ammonium compounds (QACs) will be done at six sites representing different levels of QAC pollution including textile mill effluent and sewage sludge. QAC levels will be monitored in relation to occurrence of qac genotypes, isolation media will target specific groups, and isolates characterised for diversity and presence of qac genes. A biosensor strain will be developed to assess bioavailable QAC levels in situ. Transferable resistance will be determined by exogenous isolation using seeded marked hosts, Direct community analysis will be done using degenerate sets of qac--specific PCR primers. Community DNA will also be screened for antibiotic resistance genes. Evidence for co--selection of antibiotic resistance will be sort by PCR screening and sensitivity testing of isolates.
