Abstract:
Non-thermophilic Crenarchaea have recently been detached at high levels in soil using molecular techniques, but no representative of these non-extremophile archaea has been cultured in the laboratory. Consequently, nothing is known of their physiology or metabolic activity and we are ignorant of their function in soil ecosystems. This project aims to determine their abundance, species diversity and metabolic activity in a well-characterised soil site (Sourhope), to assess the effects of soil management practices on their community structure, and to obtain non-thermophilic Crenarchaea in laboratory culture. The project will employ established molecular techniques and will also develop new techniques to relate taxonomic and functional diversity that will be of broad relevance to molecular ecological studies.
