Abstract:
Quantitative measurements will be made of rates of gene transcription in single filaments of cyanobacteria from laboratory cultures. Quantitative PCR assays will be developed for rbcS and rbcL, the genes for structural components of Rubisco. The relative transcription rates of rbc genes and genes coding for gas vesicle proteins (gvp) will be determined in Planktothrix spp. at different temperatures and during changes of temperature. The transcription rate of rbc genes will be related to the rate of photosynthesis at different temperatures and irradiances in lakes. The transcription rate of gvp genes will also be measured at different daily photon insolations, Q, and related to the photon insolation for neutral buoyancy, Qn, and the insolation at depths where these cyanobacteria stratify in lakes.
