Abstract:
The objective of the proposed research is to foster the utilisation of aDNA from charred seeds in phylogenetic and population genetic studies of cultivated plants. To achieve this objective we will test the hypotheses that: DNA survives slow charring at low temperatures in conditions of limited oxygen supply; it is possible to identify which morphologies are associated with preservation of DNA and so predict the likelihood of DNA survival on this basis; DNA decay in charred seeds has an impact on the veracity of the main PCR-based methods used to obtain variable genetic data for phylogenetic and population genetic studies; this impact is not equally severe for each method and it will be possible to identify one or more methods able to provide useable data from charred remains.
